Uncertainty of optical microscopy

Hi,
my question concerns the forum not directly:
I want to determine the uncertainty of an optical microscope according to GUM. Has anybody determined the capability of the length measurement process performed by an optical microscope?
Often pictures are taken and the length is measured in calibrated pictures so it can still be a topic of image analysis.
Perhaps anybody can give me a hint.

Sorry for the delay in reply, @pigfy. Have you considered posting this question to the confocalmicroscopy mailing list? There are many experts there who probably have opinions and/or insight about this!
