Measure object intensity distribution

Hi everyone,

I am trying to use the Measure Object Intensity Distribution module is Cell Profiler to look at a death stain distribution within spheroids (sum projected images of 3D timelapse).

Question:
Does the colour heatmap not work on an absolute scale? It changes from timepoint to timepoint and I’m having a hard time interpreting them (I have attached 2 images from consecutive timepoints to demonstrate this).

Also is my understanding of MeanFrac correct?
If the number is 1 then then intensity in this bin is what would be expected if the stain was homogeneous throughout the entire object (ex: 50% of all signal is in a bin that comprises 50% of total object area)
If >1 then there is more intensity in this bin than would be expected if the stain was homogeneous throughout the entire object.
If <1 then there is less intensity in this bin than would be expected if the stain was homogeneous throughout the entire object.

Thanks!
