Mean gray value? or use something else to analysis these two photos

Hello,

When I analysis these two IHC photos. I found the one with dark brown shape has lower mean gray value than the one without. So, when I reported my results with bar figure, it looks wired that the darker one has low number than the one without darker brown, any suggestion to improve this analysis?

Dark means small numbers, bright means large numbers, but you may invert your images, which however will also invert color, except if you use gray-value images.

how to invert color? Can I just invert the mean gray value number instead?

To invert an image choose “Edit >> Invert” of the menu of ImageJ.
