Incomplete histogram

Hi all,

I am trying to analyse images by using an image calculator (image green channel/ image red channel). the resulted image histogram is half not complete!

what does this mean?

The steps, I’ve followed :
1- subtracting the background
2- use image calculators Hepatic cells Green_1.tif (3.0 MB)

Is there any further step?

Convert your images from RGB to 8bit before processing.

You have to remove the unnecessary channels from your RGB images processing.

Here is an example as a macro (you can that also via menu).

Assuming your both example image from your original post are already open in ImageJ:

title1 = "Hepatic cells Red_1.tif";
title2 = "Hepatic cells Green_1.tif";

// select the Red image
selectWindow(title1);

// Convert RGB to R|G|B stack
run("RGB Stack");

//Delete the green and blue channel
setSlice(2);
run("Delete Slice");
setSlice(2);
run("Delete Slice");

// select the Green image
selectWindow(title2);

// Convert RGB to R|G|B stack
run("RGB Stack");

//Delete the red and blue channel
setSlice(1);
run("Delete Slice");
setSlice(2);
run("Delete Slice");

// Divide Red by Green (result as 32 bit image)
imageCalculator("Divide create 32-bit", "Hepatic cells Red_1.tif","Hepatic cells Green_1.tif");

The histogram of the result image is

But one thing about the histogram the min and max values that I have are different with large numbers, how can I change it to this range?

In Analyze>Histogram select the option ‘Use pixel value range’.
