I am trying to subtract the background (sliding paraboloid) from sum projections (32 bit) and am unsure of what radius to set after reading the ImageJ user guide.
" For 16-bit and 32-bit images with pixel value ranges different from 0–255, the radius should be inversely proportional to the pixel value range (e.g., for 16-bit images (pixel values 0–65535), typical values of the radius are around 0.2 to 5)."
So my understanding is that when subtracting the background from 16/32 bit images the object size is no longer critical? If 0.2 - 5 is typical for 16-bit then for 32-bit is the radius is even smaller?
