How to build a graph on the centers of predefined image regions and compute its edge distances? Need to label regions as vertices and to build edges between them and compute their distances. Regions are predefined based on contents of microvessel stained area and its entropy distribution parameter. Thanks.

Graphs are really a broad topic.

You can triangulate the measured regions with a delauney triangulation and then measure the distances:
