How best to quantify changes in pixel distribution?

Hello! I would like a quantitative measure of the difference between these two images. The red signal comes from mCherry-histone, and thus in image at left the chromatin is compacted more so than in the other image. How best to quantify this difference? Gracias!

I would say compacted chromatin needs less space than decompacted. Thus a good measurement would be the area which is covered by chromatin. You can measure the area by thresholding the image at a certain value and then count number of pixels.
