Cytoplasm segmentation and intensity measurement without nuclear staining

Hello! I am basically new to CellProfiler, but it does seem the best option for my images. Here is a sample_img.tif (2.0 MB) . My problem is, I have to segment cells without nuclear staining (unavailable, old images…) and measure the mean intensity in the cytoplasm.
Any suggestion or hint would be really much appreciated!

Welcome to the group!! It is very much possible to segment cells without nuclei stain. With your images also it is possible to segment then since you have reasonable signal to noise ratio. In CellProfiler you can use “IdentifyPrimaryObject” module to segment the cells. Just to start with you can look at tissue example where they done it similary for cell segmentation and even other examples might also help you.
