I am completely new for cellprofiler. I am working on a project to count the autolysosomes from the image taken by electron microscope. The autolysosomes are the medium white vacuums with clear edge in the image. The big one surrounded with black points are blood vessels. Can anybody give some ideas how to develop it?

Going from your description, I would try to find the edge and then looking for something surrounded by edges that are medium white. You might need to smooth the image before thresholding to get rid of the high frequency graininess.
Once you’ve done some of the image processing, I would take the edge enhanced image and then try to fill that in and then convert the filled in areas to object for further analysis.
Those steps should allow you to put together a pipeline.
