could you tell me what program should I use for foci coutning in yeast Saccharomyces cerevisiae?

FindFoci works very well in my experience. The software developer wrote a very thorough manual too:

Have you ever heard about The Focinator? Have you used it?

Last year I tried Focinator briefly and I don’t recall all the details but I really liked the data output you get with FindFoci. One of our microscope facility users did a more detailed comparison, including Focinator, and found FindFoci to be the closest to ground truth accuracy.
