3d image from scanning electron micrographs

Does anyone perform a 3d image from a single image obtained from scanning electron microscope? I don’t know how transform the gray level in 3d

If the gray values stand for the z-dimension you need to put the various gray value to separate slices of a stack and finally make the stack binary. For logical reasons the resulting 3D object can only be binary.

If you need this only for display in pseudo-3D, then you could use this:
https://imagej.nih.gov/ij/plugins/surface-plot-3d.html
The z dimension is the greyscale value.

Otherwise, as Herbie suggests you could do something similar to threshold decomposition, where each slice in a stack holds a range of greyscale values.
